The work is based on a rat SLK cDNA, shown to encode a protein that has tyrosine kinase activity. Several experiments have been designed to validate the cDNA and understand its biological function. First, the effect of SLK expression on ligand-induced proliferation of the transformed fibroblast cell line NRK-49F has been studied. The SLK cDNA was transfected into the cell line and found to be a potent enhancer of growth factor-induced mitogenesis. The results provide strong evidence for a role of SLK in signaling pathways, and suggest a potential role for this kinase in the growth and proliferation of mammalian cells. In order to study the function of SLK in vivo, SLK expression has been disrupted in mice. One mutant mouse line has been generated and is being characterized, and two other mutant mouse lines are planned.
